Ultimate usage not registering
I used Reyna's ultimate twice in a round, though neither of them updated the missions I have to use ultimates. The performance tab at the end of the game also showed that it had an average of 0 casts per round.
Does Reyna have a requirement such as getting a kill while in her ultimate or should I just use another agent for the quests?

if you checked the quest midgame it only updates when the game finishes
I checked at the end as well. The First blood and Play games missions went up, but the daily and weekly missions to use ultimate abilities did not.
