Thanks, does that also work if I want to store it on a different drive?

The virtual filesystem you interact with directly does not even have a concept of drives, so yes

So just to make sure I'm not messing anything up, editing this line would work? - ${BACKUP_LOCATION}: E:\Backup\immich-server\

No In your compose file you need to
${BACKUP_LOCATION}: /usr/src/app/upload/backups
${BACKUP_LOCATION}: /usr/src/app/upload/backups
and then define BACKUP_LOCATION in your .env file, or directly write
/path/to/my/backups: /usr/src/app/upload/backups
/path/to/my/backups: /usr/src/app/upload/backups

oooh I see Do I need to define the other custom locations or should I just comment them if I'm not using them or can I leave them uncommented

Locations you don't want to change should be removed or commented

Thanks Is it normal that it's been over 24h and the folder is still empty? I would assume some backup job would have run and copied data over to it by now

You need to start by selecting a folder to start the backup process, basically in your app you have to tell Immich which folder to backup. See the guide https://immich.app/docs/install/post-install#step-6---upload-your-library
